Mouse renal cell ablation and regeneration

Project Description

A specific and rapid cell ablation model mediated has been developed by my lab for nearly years. This ablation model was created by my lab via utilization of unique feature of bacterial toxin, intemedilysis (ILY)(Nat Med. 2008 Jan;14(1):98-103). ILY only lyzes human CD59 but not cells from any other species via exclusive interaction of human CD59. transgenically expressing human CD59 in mouse specific cells and injection of intemedilysin (ILY). To facilitate the utilization of this method, we have developed the cell specific ablation model by generation of Cre-inducible human CD59 transgenic mice (ihCd59) (J Clin Invest. 2016 Jun 1;126(6):2321-33.). Recently, we have crossed ihCD59 with specific renal cre line such podocyte cre line. After intermedilysin administration, ILY caused rapid and specific podocyte damages, resulting in renal failure with high protein urine, a typical feature of acute nephrotic syndrome. Currently, we are investigating the role of podocytes for maintaining pathophysiological function of kidney using this podocyte ablation model. We are studying the cellular and molecular mechanisms underlying podocyte regeration.
